Transparency and quickly turn off Aero in Vista
I think Microsoft has done a great job with the interface in Vista Aero transparency, but may not be times were you want to turn it off. Are you trying to make a screenshot and will not include online content through a window? Normal process that you have to go takes a few clicks of the mouse:
Appearance settings by clicking the Open Start ->Control Panel ->Appearance and Personalization ->Window color and appearance . Uncheck the box marked with Aero Transparency , and then press OK. Alternatively, Windows Vista Magazine is a quick guide , which will enable the option to turn off the Aero transparency in your desktop, right-click menu. There is no software to install is a simple registry hack. This makes it much easier to turn on or off switch for a moments notice.
Disabling transparency alone will probably not save a lot of system resources, since they do not close the Desktop Window Manager(DWM), but I can see how some people want to do it for a specified period.

No, the bucket makes it super pixelised and not usable... 
1) If it's a PNG file then when you import the picture in PS the transparency will be there. Therefore you will have layer with your text. Double click the text layer (in the layers tab, bottom right) and then from the dialog box opened, go to "Color Overlay", and fill it with the color you want.
2) If this won't work then: Go to Select->Color Range and click on the black text, turn the fuzziness all the way up and click ok when you are done with the selection (there should be a preview which lets you know the selected area). Now the text should be selected. From now on there are a lot of ways to fill it with color, the simplest being: choose the brush and paint the selected area
